PROBLEM TO BE SOLVED: To obtain a new nitrile hydratase having a specific amino acid sequence, expressing nitrile hydration activity, excellent in heat stability, giving in a short time host cells having nitrile hydration activity and having an activity to convert acrylonitrile into acryl amide. SOLUTION: This new nitrile hydratase is a protein consisting of two subunits which are subunit α having an amino acid sequence expressed by formula I (Xaa is defect or Met) and subunit βhaving an amino acid sequence expressed by formula II, has molecular weights measured by SDS-polyacrylamide electrophoresis 25,000±1,000 dalton for the subunit α, and 26,000+1,000 dalton for the subunit β and a hydration activity for converting acrylonitrile into acryl amide, and is excellent in heat stability. Its gene can prepare host cells having the nitrile hydration activity in a short time. The enzyme is obtained from a culture product of Bacillus smithii SC-J05-1 (FERM BP-4935) by separating and purifying the culture product. |
