PROBLEM TO BE SOLVED: To obtain both a polypeptide having a specified amino acid sequence and activities of transporting cobalt and a new gene capable of coding the polypeptide, integrated into a plasmid with a nitrile hydratase gene and providing the nitrile hydratase having high activities. SOLUTION: This new gene DNA is capable of coding a polypeptide having an amino acid sequence represented by the formula and activities of transporting cobalt. The gene DNA can be integrated into a plasmid together with a gene of a nitrile hydratase which has activities of hydrating a nitrile compound and converting the nitrile compound into an amide compound and expressed in a host cell, to thereby obtain the nitrile hydratase enzyme having high activities. The gene DNA is obtained by culturing Rhodococcus rhodochrous J1 strain (FERM BP-1478), extracting a chromosomal DNA therefrom by a conventional method, preparing a DNA library by using the extract, thereafter, screening the resultant DNA library by using a probe and collecting the objective DNA from a positive clone. |
