<p>PURPOSE:To obtain a monoclonal antibody useful for diagnosis of heart diseases and hepatic diseases, by preparing an antibody-forming cell from a mouse immunized against human LDHM subunit and cloning the antibody-forming cell. CONSTITUTION:A mouse is immunized against an isozyme of human LDH (lactic dehydrogenase) V type, a splenocyte is collected from the mouse, fused to a mouse myeloma cell, a cell group to produce an antibody is selected by ELISA and cloned to give fused cell LD-10, 11, 17, 21, 25, 52, 58 and 66 strains to produce a monoclonal specific to human LDHM subunit. The strain is cultivated in a medium or transplanted in abdominal cavity of a mouse, made into ascites tumor and the aimed monoclonal antibody is obtained from the culture solution or ascites.</p> |
