PURPOSE:To profitably provide the subject compound useful as a thrombosis remedy, etc., by combining the DNA sequence of a human tissue type plasminogen-activating factor with a manifestation vector, inserting the combined vector into a host cell and subsequently culturing the host cell while the osmotic pressure of the medium is adjusted with bicarbonate ions. CONSTITUTION:The DNA sequence of a human tissue type plasminogen- activating factor(tPA) is bonded to an adequate promoter and the bonded sequence is combined with a manifestation vector. The prepared vector is inserted into a host cell to transform the cell and the transformed cell is cultured in a medium using bicarbonate ions to adjust the osmotic pressure of the medium to >=350 miliosmol/l to produce the tPA, followed by purifying the tPA stored in the medium with an affinity column and a gel filtration column, etc., to provide the objective tPA. The concentration of the bicarbonate ions in the medium is preferably >=3g/l. |
