PROBLEM TO BE SOLVED: To simply detect a DNA mutation useful for detecting gene deficiency, diagnosis, etc., of cancer with high sensitivity by carrying out the polymerase chain reaction (PCR) of a sample with a labeled primer and then performing the smoothing treatment of the 3'-terminal side of the resultant PCR product. SOLUTION: The polymerase chain reactional (PCR) method is carried out for a sample containing a gene by using a fluorescence-labeled oligonucleotide primer to replicate a DNA in a gene region where a gene polymorphism is present from the gene in the sample. The smoothing treatment of the 3'-terminal side of a DNA fragment of the resultant replicated PCR product is carried out to analyze the prepared DNA fragment according to a single-stranded conformation polymorphism method (SSCP method). Thereby, the loss of heterozygosity (LOH) is detected to simply detect the DNA mutation used for diagnosis, etc., of cancer by the presence or absence of the sensitivity to a restriction enzyme HaeIII in a p53 tumor suppressor gene. |
