PROBLEM TO BE SOLVED: To provide an efficient cloning process for a DNA coding the substrate protein of a protein kinase. SOLUTION: The host in to which a DNA inserted into an expression vector is transduced is plate-cultured. A membrane is brought into contact with the plate to transcript the expressed protein from the plate to the membrane. The membrane is peeled off from the plate, a labeled phosphate donor and a protein kinase are added to the membrane to phosphorylate the expressed protein and to detect the phosphate label bound to the expressed protein. Finally, the DNA is isolated from the clone on the plate corresponding to the positive site on the membrane. |
