<p>The invention provides a method for measuring the amounts of oligonucleotides, particularly applicable to monitoring the pharmacokinetics of antisense oligonucleotide pharmaceuticals, which permits resolution of homologous oligonucleotides differing by a single terminal nucleotide. The method consists in the ligation of the target oligonucleotide to a (labeled) primer with the help of a bridging oligonucleotide hybridizing to the ends of both target and primer.</p> |
