PROBLEM TO BE SOLVED: To obtain the subject hydrase reacting with chondroitin sulfate at a specific acidic pH, not reacting with hyaluronic acid, keratan sulfate, heparan sulfate and heparin and useful as a treating agent for hernia of the intervertebral disk and a reagent, etc., for research of sugar chain. SOLUTION: This new chondroitin sulfate hydrolase has action hydrolyzing N-acetylhexosaminide bond of glucosaminoglycan and reacts with chondroitin sulfate at pH 5 and does not react with hyaluronic acid, keratan sulfate, heparan sulfate and heparin and reacts with chondroitin sulfate and hyaluronic acid at pH 3.5 and has optimum pH in the neighborhood of pH 5 (using chondroitin sulfate as a substrate and using 50mM citric acid-Na2 HPO4 buffer solution containing 0.15 M NaCl as a buffer solution at 37 deg.C) and has isoelectric point in the neighborhood of pH 5 and it is expected that the hydrolase is utilized as a treating agent for hernia of the intervertebral disk and a reagent, etc., for research of sugar chain. |
