PROBLEM TO BE SOLVED: To provide a method for detecting ligand in an organism sample for utilizing alkali phosphatase which is highly pure, is stable, and has a specific activity which is nearly equal to enzyme originated from the small intestine of calf, its reagent, a method for determining ligand in a sample, and a method for determining the nucleic acid sequence in the sample. SOLUTION: Ligand in an organism sample is detected by utilizing alkali phosphatase with following scientific and chemical properties (1)-(6), the ligand in the sample is determined, and at the same time the nucleic acid sequence in the sample is determined, namely (1) catalysis reaction: orthophosphoric acid monoester +H2 O→alcohol + orthophosphoric acid, (2) activation and stabilization agent: Mg<2+> and Co<2+> ,(3) thermal stability: stable for at least three minutes by a treatment of pH7.5 and 60 deg.C, (4) specific activity: at least 2,300U/mg, (5) no sugar chain, and (6) molecular weight: 140,000-150,000 (gel filtering method) 65,000-67,000 (SDS-PAGE).
