PROBLEM TO BE SOLVED: To efficiently form the polymer of microgene, useful for creating various functional proteins, etc., by adding different DNAs to both the ends of a microgene fragment, and subsequently subjecting the prepared DNA containing complementary sequences to a ligase reaction. SOLUTION: A specific DNA fragment (A) expressed by formula I, etc., and a specific DNA fragment (B) expressed by formula II, etc., are addition- reacted to the ends of a microgene fragment, respectively, to prepare the product containing the DNA sequences (a) and (b) comprising the DNA sequences (A) and (B) at least containing complementary sequences, respectively. The product is converted into a single-strand DNA wherein the DNA sequences (a) and (b) are connected to each other, which is subjected to the ligase reaction of the microgene to obtain the objective random polymer of the microgene. The random polymer can be applied to produce the gene library necessary for the creation of various functional proteins, to create functional proteins large in the molecular weights, and to develop new physiologically active substances and polymers. |
