The invention relates to a reagent for the in vitro diagnosis of the infectious mononucleosis based on bound horse erythrocytes, characterized by a binding degree sufficient to prevent any "agglutination on plate" of said erytrhocytes, when a suspension of said erytrhocytes in an aqueous vehicle is brought in contact with a sufficiently diluted human serum, for example diluted to 1/40, but authorising a "mat-pattern agglutination", particularly in a tube having an approximately conical or U-shape and, or in the microtubes of tubules of a microplate. It also relates to an in vitro diagnosis method characterized by contacting said reagent with the biological sample to be tested, which is itself diluted, and the detection of a mat-pattern micro-agglutination when the sample contains antibodies which are characteristic of the infectious mononucleosis.
