PROBLEM TO BE SOLVED: To obtain the platelet factor useful e.g. for the neutralization of heparin from an easily available material with a simple operation in high yield by contacting a solution containing the platelet factor 4 with chitin sulfate to effect the adsorption of the factor and eluting the factor from the adsorbent. SOLUTION: The objective platelet factor 4 useful as a heparin neutralizing agent, malignant tumor suppressing agent, etc. is produced in high yield by centrifuging a collected human blood to remove plasma, adding 0.155M ammonium chloride solution to the remaining hemocyte component, leaving the mixture at rest for 10min at room temperature, subjecting to centrifugal separation, removing precipitated leukocyte, subjecting the supernatant to centrifugal separation at a higher speed, collecting the precipitated platelet, freezing and melting the platelet, mixing with a 15mM tris-HCl buffer solution (pH7.6), subjecting to centrifugal separation, collecting the supernatant as a platelet extraction liquid, adding sodium chloride to the supernatant to attain a concentration of 0.4M, contacting with chitin sulfate to effect the adsorption of platelet factor 4 and eluting the adsorbed substance with 1.0M sodium chloride solution. |
