摘要 <p>Polynucleotides (particularly DNA) in cell lysate are separated from proteins by reacting the proteins with a cross-linked silica material having free CHO or CO groups, so as to remove the proteins as a solid or semi-solid material in a two-phase solvent system. The lower phase is chloroform and the upper aqueous phase contains the DNA; and the protein-containing silica is present as an intermediate solid or semi-solid layer. The cross-linked silica is preferably prepared by cross-linking silica gel of particle size 250-400 mesh with 3-aminopropyltriethoxysilane, followed by reaction with gluteraldehyde to introduce protein-reactive CHO groups.</p>
