RECOVERY OF INGREDIENT EXTRACTED FROM MACROPHAGE

PROBLEM TO BE SOLVED: To simply and massively obtain the subject ingredient useful as an additive for cell culture media, etc., by suspending a bovine lung in a saline solution having a specific salt concentration, removing dissolved substances, suspending the residue in a saline solution having a lower salt concentration than the above saline solution and subsequently recovering the dissolved substance. SOLUTION: This method for recovering an ingredient extracted from macrophage comprises slicing a bovine lung, suspending the slices in a 0.5-0.7wt.% saline solution, leaving the suspension at 4 deg.C for 20min, separating off the supernatant by a decantation method to remove the dissolved substances, recovering the precipitate to recover erythrocytes from the slices of the bovine lung and simultaneously elute erythrocyte intracellular ingredients into the suspension, suspending the recovered precipitate in a <0.5wt.% of saline solution, leaving the suspension at 4 deg.C for 30min, separating the suspension into the supernatant and the precipitate by a decantation method, collecting the supernatant to obtain the macrophage extraction solution containing macrophage intracellular ingredients, and subsequently lyophilizing the extraction solution. The obtained objective macrophage extraction ingredient is useful as an additive for media used for culturing cells.
