A novel circular DNA molecule (I), useful in gene therapy, comprises >= 1 sequence of interest, the replication of which is controlled by an origin of replication (ORI) from a plasmid or bacteriophage, which is only active in a host cell in the presence of one or more specific proteins foreign to that cell.
