FIELD: biotechnology, biochemistry, bacteriology. SUBSTANCE: biomass is collected at log-phase growth at minimal sporulation of bacterial culture. Biomass is treated with lysozyme in the presence of EDTA at concentration 20 mg/1 g biomass. Obtained suspension is frozen three-fold in liquid nitrogen followed by thawing at 60 C. Freezing time is 40-60 s. Then suspension is subjected for additional analysis with buffer tris-sodium-dodecyl sulfate (tris-SDS). Nucleic acids were purified with phenol and precipitated with cold ethanol. Method provides the preparing large microbe cells amount, their disinfection. Nucleic acid yield is increased by above 70% as compared with prototype. EFFECT: improved method of preparing, increased yield. |
