PROBLEM TO BE SOLVED: To obtain a new human antithrombin III mutant, having high anticoag ulant activities in the absence of a heparin and useful as a therapeutic agent, etc., for thrombotic diseases by adding a peptide containing a tyrosine undergo ing the sulfating modification to the C-terminal of a human antithrombin III (AT III). SOLUTION: This new AT III mutant comprises a substance containing a part or all of a protein, represented by formulas I to III and prepared by adding a peptide containing a tyrosine undergoing the sulfating modification of the C-terminal of a human antithrombin III (AT III). The mutant has high anticoagulant activities in the absence of a heparin and is useful as a therapeutic agent, etc., for thrombotic diseases. This new AT III mutant is obtained by using a human liver cDNA library, carrying out the cloning with a synthetic oligonucleotide as a probe, preparing a cDNA coding for the AT III, joining a DNA coding for the peptide containing the tyrosine represented by formulas I to III to the C-terminal side thereof, integrating the resultant DNA into a vector, transducing the prepared vector into a hose cell, culturing the host cell and expressing the gene. |
