Isolated DNA from Streptomyces achormogenes encoding SacI restriction endonuclease is new. Also claimed are: (1) a recombinant DNA vector contg. a DNA segment encoding SacI; (2) an isolated DNA from ATCC 69767 encoding SacI restriction endonuclease and methylase; (3) a cloning vector comprising the DNA of (2); and (4) a host cell transformed with (1) or (3).
