NEW MODIFIED NUCLEOTIDE AND ITS PRODUCTION AND PRODUCTION OLIGONUCLEOTIDE USING THE SAME

PROBLEM TO BE SOLVED: To obtain a new modified nucleotide for synthesis, etc., of medicines and agrochemicals, capable of readily carrying out deprotection even if chain length becomes long in synthesis of oligo-RNA by reacting a new 2-(levulinyloxymethyl)benzoic acid with a nucleoside derivative. SOLUTION: This new 5'-O- 2-(levulinyloxymethyl)benzoyl}nucleoside derivative is expressed by formula I (B is a nucleic acid base or a nucleic acid base in which a protective group is introduced; R is H or hydroxyl group in which a protective group is introduced). A new compound 2-(levulinyloxymethyl)benzoic acid expressed by formula II (Me is methyl) is used as a protecting group of nucleoside in synthesis of oligonucleosides. The compound has no influence on the condensation ratio of RNA oligomer chain elongation reaction due to bulkiness and removal of the protective group is readily carried out even if chain length becomes long. The modified nucleoside is obtained by reacting 2-(levulinyloxymethyl)benzoic acid (salt) of formula II with a nucleoside derivative having formula III.
