PROBLEM TO BE SOLVED: To obtain a new amylase acting on polysaccharides and oligosaccharides to isolate glucose and useful as an enzyme for food, an enzyme for pharmaceutical use, etc., by culturing a microorganism belonging to the genus Acremonium in a nutrient medium and collecting the culture product. SOLUTION: A microbial strain belonging to the genus Acremonium [e.g. Acremonium sp. YT-78 (FERM P-15097)] is cultured in a nutrient medium and the accumulated product is separated to obtain the objective new amylase acting on polysaccharides and oligosaccharides to isolate glucose, acting on starch, glycogen, amylose, amylopectin, maltoheptose, maltohexose, maltopentose, pullulan, maltose and panose, scarcely acting on isomaltose, dextrin and cyclodextrin, having an optimum pH of about 5.0, an optimum temperature of about 50 deg.C, stable at pH 5-9 or thereabout (40 deg.C, 30min), stable at 40 deg.C for 30min (pH 5.0) and having a molecular weight of about 74,000 (SDS-PAGE). |
