PROBLEM TO BE SOLVED: To obtain the subject new enzyme by culturing a bacterium belonging to the genus Bacillus, capable of producing maltose phosphorylase, having thermostability, carrying out enzyme reaction at a high reaction temperature, reducing contamination with various germs, providing trehalose in a short time. SOLUTION: This new enzyme is obtained by culturing a bacterium belonging to the genus Bacillus [e.g. Bacillus sp.MK-1 (FERM P-15,045) etc.], capable of producing maltose phosphorylase, in a nutrient medium and collecting the product from the culture product, provides equimolar amounts of glucose and β-glucose-1-phosphate when treated with maltase in the presence of phosphoric acid, forms equimolar amounts of maltose and phosphoric acid when treated with glucose and β-glucose-1-phosphate, has activity by treatment in 10mM acetic acid buffer solution (pH6.0) at 60 deg.C for 15 minutes, equal to or higher than 80% activity of nontreatment, 55-70 deg.C optimum temperature, optimum pH6.0 to pH7.0, 150,000-190,000 molecular weight measured by gel filtration and industrially and advantageously provides β-glucose-1-phosphate, trehalose, etc. |
