PROBLEM TO BE SOLVED: To proliferate a base sequence by specifically cyclizing a base sequence to be detected by ligase chain reaction using a probe in which the base sequence to be detected and its complementary chain are adjacent and to which probes to be bound by ligase reaction are bound. SOLUTION: In proliferating a sequence A1A2 comprising continuing base sequences A1 and A2 in a single strand nucleic acid to be detected, a hybrid is formed between a probe B obtained by binding a nucleic acid comprising a sequence B1 complementary to the sequence A1 to a nucleic acid comprising a sequence B2 complementary to the sequence A2 and the sequence A1A2 in the nucleic acid, and 5' end of the sequence B1 of the probe B is bound to 3' end of the sequence B2 by ligase and the hybrided double strand is thermally denatured, and a hybrid is formed between the thermally denatured nucleic acid complex and the probe B or a probe A obtained by connecting the nucleic acid comprising the sequence A1 to the nucleic acid comprising the sequence A2, and 5' end of the sequence B1 of the probe B is bound to 3' end of the sequence B2 by the ligase and the base sequence to be detected is proliferated by ligase chain reaction. |
