PROBLEM TO BE SOLVED: To obtain a fission yeast polyploid strain simply, accurately and in high frequency by treating a fission yeast such as Schizosaccharomyces pombe under pressurization, culturing the fission yeast on an agar medium containing a pigment and selecting polyploid colonies based on a color change. SOLUTION: This harvest of a fission yeast polyploid strain comprises inoculating the fission yeast such as Schizosaccharomyces pombe JY1 strain to a medium, shaking the medium at 30 deg.C for 30 hours, centrifuging the culture solution to collect the yeast, washing the yeast with 0.86% NaCl aqueous solution, suspending the yeast in 0.86% NaCl aqueous solution, enclosing the yeast suspension in a polyethylene container, treating the yeast under 100-250Mpa hydrostatic pressure at 25 deg.C for 10 minutes using a pressure treating device, culturing the fission yeast after the heat treatment on an agar medium containing a pigment and selecting the polyploid colonies based on a color change. Thus, a screening can be carried out simply and accurately and the polyploid strain of the objective fission yeast can be effectively obtained in an extremely high frequency. |
