PURPOSE: To obtain a new protein having a specific amino acid sequence, having an ubiquitin-bound enzymatic activity playing an important role in an intracellular protein-decomposing system, capable of being produced in a large amount by a genetic recombination technique, and useful as a reagent for the research of ubiquitin. CONSTITUTION: This new protein comprises a human ubiquitin-bound enzyme E2 containing an amino acid sequence of the formula, having an ubiquitin-bound enzyme activity playing an important role in an intracellular protein decomposition system, and useful as a reagent for researching the ubiquitin, etc. The protein is obtained by culturing a human epidermal cancer cell strain, separating mRNA by a conventional method, synthesizing a cDNA in the presence of the separated mRNA as a template, producing a cDNA library by a conventional method, determining the base sequence of an arbitrarily selected clone, detrieving an amino acid sequence coded in the base sequence from a protein base, screening a clone containing the ubiquitin-bound enzyme cDNA, recovering the cDNA, inserting the recovered cDNA into an expression vector, and subsequently expressing the protein in a host cell. |
