FIELD: medicine; differential diagnostics of different pathological conditions of organism. SUBSTANCE: method involves tissue sample fixation, dehydration, clarification and paraffinization, performed in home microwave stove at output power of 450 W; fixation is realized within 20 seconds period at power of stove setting at 90% of maximum; after draining off fixative tissue sample, refilled with 96%-alcohol, is placed in microwave stove for 10 minutes at power of stove adjusted at 20-30% of maximum, with sample container being additionally surrounded from four sides by water loading, which full volume is equal to 1000 milliliters; dehydration is performed three times under similar condition of microwave stove; then tissue sample is filled with mixture of alcohol and chloroform in respect of 1:1, and placed in microwave stove for 5 minutes at power of stove setting at 30% of maximum, herewith water loading of full volume about 1250 ml is spread among five containers arranged on circumference of sample container at optimal radius of 3 cm; then sample container refilled with pure chloroform is placed again into microwave stove for 10 minutes at power setting at 70% of maximum; paraffinization is performed according to routine procedure by filling tissue sample with mixture of chloroform and paraffin and placing sample in microwave stove for 5 minutes at power adjusted at 50% of maximum, under condition of preceding water loading; then sample is filled with melted paraffin according to routine procedure and placed again in microwave stove for 15 minutes at power setting at 40% of maximum, under water loading similar to that of preceding stage. EFFECT: reduction of time, required for manufacturing histological preparations, from 10-12 days to 1 hour 20 minutes at maintenance of high quality of preparations. |
