FIELD: biotechnology. SUBSTANCE: new microbe enzyme preparation is proposed. Properties: total proteolytic activity is 300 U/g (by Kunitz method) or 300000 U/g (by tyrosine method); preparation hydrolyzes casein, isoelectric point is 5.9 and 6.0, pH optimum is 6.8-7.8, pH range of stability is 6.8-7.8; thermostability: activity is decreased above 45 C; Michaelis constant = 0.9 x $$$ M; substrate: sodium caseinate; inhibitors: EDTA, phenylmethylsulfonylfluoride, o-phenanthroline (at concentration above $$$ M). Its activity exceeds that of alkaline protease from Bacillus subtilis but, in contrast to the latter, makes both depilation and softening that excepts pancreatin use, decreases consumption of sodium sulfide (polluting agent of environment) by 2-3-fold, increases yield of ready leather up to 5%. Enzyme preparation ("ГЗХ"-form) is obtained from cultural fluid filtrate of the strain Streptomyces lavendulae "ИНМИ"-36-SV by drying in spraying drier, at 120-125 C at inlet and at 60-65 C at outlet and addition of 1% NaCl to dry powder. EFFECT: enhanced quality and effectiveness of enzyme preparation. |
