<p>PURPOSE: To efficiently obtain the subject cell line by fusing a myeloma cell with the lienal cell of a mammalian immunized with human villous gonadotrophin, testing the binding ability of the fused cell to hormones to screen the objective cell, and subsequently cloning the fused cell. CONSTITUTION: A method for producing a monoclonal antibody-producing cell line by fusing a cell line (e.g. 3×63-Ag 8.653 originating from a mouse myeloma) originating from a mammalian with the lienal cell of a mammalian (e.g. mouse) immunized with villous gonadotrophin originating from a man, and subsequently cloning the fused cell, the improvement comprises testing the binding ability of the fused cell to hCG, theα-subunit (hCG-α) of the hCG, theβ-subunit of the hCG, luteal hormone(LH), follicular hormone(FSH), and thyroid-stimulating hormone(TSH) by an immunoassay method before the cloning process to screen the objective cell. Thus, the objective monoclonal antibody-producing cell line for producing the anti-hCG monoclonal antibody for pregnancy diagnosis, etc., is obtained.</p> |
