<p>PURPOSE: To obtain a new monoclonal antibody, capable of specifically recognizing a C-terminal peptide in a human I type procollagen and useful for immunoassay, etc., of the amount of the C-terminal peptide in the human I type procollagen in a sample such as a humor and useful for diagnosis, etc., of hepatic fibrosis. CONSTITUTION: This monoclonal antibody is capable of specifically recognizing a C-terminal peptide in a human I type procollagen, further immunologically and rapidly carrying out the determination of the C-terminal peptide in the human I type procollagen in a sample such as a humor with high sensitivity and useful for diagnosis or treatment, etc., of various fibroses such as hepatic fibrosis. The monoclonal antibody is obtained by culturing a fibroblast derived from a human fetal lung in a culture medium, extracting a purified human I type procollagen from the resultant cultured product, administering the resultant procollagen together with an adjuvant to a Blab/c mouse, immunizing the mouse therewith, collecting a cell of the spleen from the mouse, fusing the cell to a cell of a myeloma, screening the prepared hybridoma, selecting a strain capable of producing the antibody, cloning the selected strain and culturing the cloned strain.</p> |
