PURPOSE: To obtain a new enzyme derived from Fungi Imperfecti Colletotrichum lindemuthianum, acting on N-acetylglucosamine-containing substrate, eliminating N-acetyl group, capable of producing a chitosan oligomer useful in the field of foods, etc. CONSTITUTION: This new chitin deacetylating enzyme is derived from Fungi Imperfecti Colletotrichum lindemuthianum (ATCC 56,676), acts on a substrate containing N-acetylglucosamine residue to eliminate N-acetyl group, is stable at pH 6 to pH 10.5, optimum pH 11 to pH 12 at 30 deg.C, 60 deg.C optimum temperature at pH 8.5, about 31,500 molecular weight by SDS-PAGE, about 33,000 by gel filtration method, about 3.7 isoelectric point pI and an N end amino acid sequence of the formula and is useful for producing a chitosan oligomer useful in the field of foods. This enzyme is obtained by culturing Colletotrichum lindemuthianum, in a medium, filtering a cell culture solution, adding ammonium sulfate to the filtrate so as to give a 80% saturated solution and purifying formed precipitate by chromatography. |
