PURPOSE: To obtain the subject new enzyme from a fungus belonging to the Cladosporium, capable of producing an N-alkylglycine oxidase, useful for measuring Nε-carboxymethyllysine in a body fluid as judging criteria for diagnosing and treating diabetes, etc. CONSTITUTION: This new N-alkylglycine oxidase catalyzes a reaction of the formula (R is an alkyl group or its derivative) to oxidize an N-alkylglycine in the presence of an enzyme to form an alkylamine, glyoxylic acid and hydrogen peroxide, specifically acts on sarcosine, N-ethylglycine, N-carboxymethyl-6- aminocaproic acid and Nε-carboxymethyllysine and has optimum pH 8.0 to pH 10.0, a stable pH range of pH 7.5 to pH 9.5 by treatment at 30 deg.C for 10 minutes and about 45,000 molecular weight (gel permeation method). The enzyme is obtained by culturing a strain [e.g. Cladosporium sp.G-10 (FERM P-14,887), etc.] belonging to the genus Cladosporium in a medium. |
