1290277 5 - Phenyl - 1,9 - alkylene - 1,4- benzodiazepin-2(3H)-ones A H ROBINS CO Inc 19 April 1971 [14 April 1970] 26652/71 Heading C2C Novel 5 - phenyl - 1,9 - alkylene - 1,4 - benzodiazepin-2(3H)-ones of the general formula wherein R is a hydrogen or chlorine atom and n is 1, 2 or 3, having anticonvulsant activity, are prepared by reaction of an N-bromo-acetyl compound of the general formula with ammonia in an alkanol solvent or with an alkyl glycinate. N-bromoacetyl compounds of the second general formula above are prepared by reaction of a ketone of the general formula with bromoacetyl bromide. Ketones of the third general formula above are prepared (a) by reaction of a heterocyclic amine of the general formula with oxalyl chloride, treatment of the resulting acid chloride of the general formula with aluminium chloride, with heating, followed by hydrolysis with ice-water (when the acid chloride is not spontaneously cyclized), oxidation of the resulting dione of the general formula with hydrogen peroxide in a basic aqueous solution and reaction of the resulting carboxylic acid with phenyllithium; (b) treatment of a heterocyclic amine of the fourth general formula above with sodium nitrate, conc. HCl and ice, treatment of the resulting nitroso compound of the general formula with zinc and acetic acid, reaction of the resulting amine of the general formula with phenylacetone and sodium acetate in acetic acid, and treatment of the resulting heterocyclic amine of the general formula with ozone in acetic acid; and (c) reaction of a ketone of the general formula with benzoyl chloride in the presence of zinc chloride, followed by acid hydrolysis. |
