<p>PURPOSE: To obtain a monoclonal antibody which binds to a fragment of bovine or human lactoferrin, but not to natural lactoferrins themselves and can determine the antimicrobial peptide of a human or bovine source lactoferrin fragment with high sensitivity. CONSTITUTION: This novel monoclonal antibody which binds to antimicrobial peptides of formulas I and II as fragments of human and bovine lactoferrins but does not bind to natural lactoferrins. The antibody is useful for high- specificity and high-sensitivity detection and determination of an antimicrobial peptide originating from lactoferrins contained in gastric juice, intestinal contents, feces, blood, urine and the like in no need of complicated equipments, etc. The monoclonal antibody is obtained by dosing to BALB/c mice a complex of the bovine or human lactoferrin fragment with a carrier protein for immunization, collecting spleen cells to fuse them with myeloma cells, subjecting the fused cells to selection culture to select a strain producing the antibody, obtaining the hybridoma by cloning and culturing the hybridoma.</p> |
