PROBLEM TO BE SOLVED: To obtain an oligonucleotide labeling kit which contains a primer labeled by a group of several kinds of coloring agents or fluorescent coloring agents by using the group of the coloring agents or the fluorescent coloring agents whose absorption or fluorescence is maximum. SOLUTION: A group of four kinds of coloring agents or fluorescent coloring agents whose absorption or fluorescence is differently maximum are used. Thereby, it is not required that fragment groups generated in four kinds of synthetic reactions are applied to separate gel tracks-as different from a prior art method for a DNA sequencing. Their labeling operation is chemically bonded respectively to primers, and the synthesis of fragment strands is started by using them. Then, respective labeled primers are combined with one kind of a deoxynucleotide, and they are used for a prime synthetic reaction by the Klenow fragment of a DNA polymerase. At this time, four kinds of synthesized oligonucleotide primers satisfy conditions that a characteristic as a primer, e.g. a free 3' hydroxyl group used to extend a polymerase chain is provided and that it is complementary with the peculiar region of the 3' hydroxyl group of a cloning insert. |
