PURPOSE: To obtain the new antitumor active substance GIF having a specific amino acid sequence, having an action for specifically inhibiting the multiplication of tumor cells against the tumor cells, and useful as an antitumor agent, etc., by culturing immune-charging cells originated from a mammalian to induce and produce the substance. CONSTITUTION: An antitumor activity substance GIF contains an amino acid sequence of the formula as its constituting protein, exhibits a single protein absorbance peak coincident with the activity of' the GIF in a negative phase high performance liquid chromatography, is migrated as a single band at the position of a mol.wt. of 17-18K in a SDS-polyacrylamide gel electrophoresis (SDS-PAGE) in the presence of 2-mercaptoethanol, exhibits an isoelectric point (PI) of approximately 6.5-7.02 in an isoelectric point electrophoresis (IEF) and has a tumor cell multiplication-inhibiting activity. The GIF is obtained by culturing human peripheral blood lymphocytes in a medium containing an antitumor substance-inducing agent, kidney bean lectin, at 37 deg.C for 3 days in a carbon dioxide culture device, separating the product from the supernatant of the culture solution as a raw solution material, and subsequently purifying the product. |
