<p>This invention relates to methods and compositions comprising nicotinylalanine, and/or related analogues, and an inhibitor, such as aspirin, of glycine conjugation, which are useful for inhibiting cellular poly-(ADP-ribose) synthetase in vitro and/or in vivo. This enzyme is activated in a variety of toxic and pathological conditions and is inhibited by nicotinamide. B6 may also be present in the compositions of this invention in place of, or in addition to the inhibitor of glycine conjugation. Such pathological conditions include, for example, neurodegenerative disorders, viral infections, autoimmune diseases and cancer. Accordingly, this invention relates to methods of reducing cellular toxicity and treating such diseases by increasing cellular nicotinamide using the methods and compositions of this invention.</p> |
