PURPOSE: To provide a peptide bond-hydrolyzing agent which contains o- phenanthroline and a copper complex and can hydrolyze a compound having peptide bonds such as protein or peptide to give a hyhydrolyzate without using acid or protease. CONSTITUTION: o-Phenanthroline, copper sulfate andβ-mercaptoethanol are dissolved in 10mM acetate buffer solution (pH 6) andα-casein is admixed thereto with stirring for 1hour at 37 deg.C to give this peptide bond-hydrolyzing agent which can efficiently produce the peptide hydrolyzate from protein or peptide using no acid or protease.
