FIELD: medicine, dermatology. SUBSTANCE: preparations of non-immobilized tissue slices 5 mcm thick prepared in cryostate (minus 20 C) are treated with 40-60 % aqueous solution of ethanol, washed with physiological solution (pH 7.0-7.4) for 10 minutes, treated with luminescent anti-Ig serum for 30 minutes, washed for 10 minutes and placed it into 60-% neutral glycerol. The treatment of sections for 1-3 min with 40-60% aqueous spirit shows that such mild denaturating agent provides stable antigen-antibody bond, prevents their carrying away from intercellular adhesive substance. For the treatment there were used 10-%,20-%,30-%,40-%,50-%, 60-%, 70-%,80-% and 96% aqueous mixture of ethanol. Optimum mixture is 50-% one. EFFECT: increased accuracy of pemphigus vulgaris identification. |
