PURPOSE: To obtain a midkine protein (MK) useful e.g. as an agent for the restoration and regeneration of tissue by selecting a vector transporting a cDNA having a DNA sequence of MK gene, inserting the vector into a vector and introducing into a cell to effect the expression of the gene. CONSTITUTION: This midkine protein useful as a treating agent and a wound- healing agent capable of promoting the restoration and regeneration of the damaged or destructed tissue such as organ or vessel in the living body is produced in high efficiency by selecting a virus such as baculovirus having a polyhedrin gene promoter characteristic as a vector for transporting a cDNA having a nucleotide sequence corresponding to midkine protein (MK) gene, inserting the DNA of the virus into the vector, introducing the vector into a cultured cell such as an insect cell (e.g. Sporoptera-brugiperda cell) to produce an expression system for transporting MK, culturing the cell, fractionating the MK from the cell with a carrier and separating and purifying the obtained MK. |
