PURPOSE: To provide a method for attaching biomolecules having carboxyl groups to an aminated solid surface. CONSTITUTION: Biomolecules are reacted with carbodiimide to activate the carboxyl groups of the biomolecules, and the carbodiimideactivated biomolecules are reacted with a solid surface to cause the aminated solid surface to covalently attach to the biomolecules thereby selectively restoring the carboxyl group functionality to the biomolecules. The selective restoration of the carboxyl group functionality is carried by weak hydrolysis to restore the functionality of the biomolecules. Since the chemical bonds by the carboxyl groups between the biomolecules and the aminated solid surface remain, the process of restoring the carbonyl group functionality is selective. |
