PURPOSE: To obtain a variation type human tumor necrosis factor useful as a carcinostatic agent, effective for various tumor cells, low in adverse effects, having a specific amino acid sequence prepared by replacing the second Arg of a human tumor necrosis factor with Lys. CONSTITUTION: This variation type human tumor necrosis factor has an amino acid sequence containing an amino acid sequence of the formula, which is obtained by replacing the second arginine of an amino acid sequence of a human tumor necrosis factor with lysine, has high antitumor effect on various tumor cells, only slight toxicity and is useful as an active ingredient for a carcinostatic agent. The variation type human tumor necrosis factor is obtained by synthesizing a base sequence in which a codon corresponding to arginine as the second amino acid residue is replaced with a codon corresponding to lysine, in a 157 amino acid sequence of natural type human tumor necrosis factor-α, amplifying the base sequence by a PCR method, incorporating the amplified base sequence to a plasmid vector, transducing it into a host such as Escherichia coli and manifesting the gene. |
