PURPOSE: To specifically detect the gene by recovering RNA from a specimen, holding at a specific temperature in the presence of a reverse transcriptase and a specific primer complimentary to the RNA virus gene, synthesizing cDNA, amplifying and analyzing. CONSTITUTION: An RNA is recovered from a specimen and a resultant reacting solution is kept at a temperature preventing bonding to a sequence other than a complimentary sequence to the following primer until initiation of the reaction, in the presence of a reverse transcriptase and each antisense primer or sense primer having a base number of at least 15 capable of hybridizing to the RNA virus plus strand or minus strand gene, and a cDNA is synthesized from the RNA at a temperature not damaging enzyme activity of the reverse transcriptase, the DNA is amplified by subjecting resultant cDNA to a PCR reaction, the PCR product is analyzed and respective of the plus strand or the minus strand to specifically detect RNA virus plus strand or minus strand gene in the specimen. |
