发明名称 METHOD FOR SEPARATION OF HIGHLY DISPERSED FRAGMENTS OF PURPURIC MEMBRANES CONTAINING BACTERIORHODOPSIN
摘要 FIELD: microbiology; methods for separation of proteis, particularly, bacteriorhodopsin contained in purpuric membranes of microorganisms of species HALOBACTERIUM HALOBIUM; may be used in biotechnology and photochemical and electronic industries, scientific research practice in study of photo- and electrochromic conversions, and also for preparation of biologic photorecording material of repeated use. SUBSTANCE: method for production of purpuric membrane fragments sizing from 50 to 400 nm and containing bacteriorhodopsin provides for separation of purpuric membranes from biomass of hallophyllic bacteria by the method of cyclic extrusion at pressure of 150 MPa in 1 to 5 cycles and subsequent procedure of centrifuging and washing out in bidistilled water with 25000-100000 g for 30 min, each procedure is continued until relation of optical densities of suspension of purpuric membrane fragments with wave lengths of 280 nm and 560 nm becomes equal to 1.9 to 2.1, In so doing, a version is possible when procedure of centrifuging and washing in bidistilled water is accomplished in two stages, first with 25000 g for 30 min and then, with 100000 g for 30 min, alternately. Prior to disintegration biomass is placed in bidistillate in concentration about 100 wt.-%, and after mixing is passed through homogenizing valve of special design at invariable preset pressure. Bacteriorhodopsin yield is not less than 1 g from 100 g of biomass, or 40 l of cultural suspension of cells of halobasteria. EFFECT: increased efficiency of the process with simultaneous lowering of price; provision of regulated reduction of average size of particles of fragment of purpuric membranes containing bacteriorhodopsin and standard deviation of their distribution in sizes.
