PURPOSE: To rapidly test the sensitivity of an acid-fast bacterium to an antituberculosis medicine by treating the acid-fast bacterium in a culture medium containing the antituberculosis medicine at a prescribed temperature for a prescribed time, extracting RNA and subsequently analyzing the RNA. CONSTITUTION: An acid-fast bacterium in a culture medium containing an antituberculosis medicine such as rifampicin is treated for a time and at a temperature for lowering or raising the level of 71KDa heat shock protein(HSP) mRNA in response to whether the acid-fast bacterium is sensitive or resistant to the antituberculosis medicine. RNA is extracted from the treated acid-fast bacterium, and HSPmRNA is detected. The sensitivity test of the acid-fast bacterium to the antituberculosis medicine can be shortened to a few days, and further fast reflected to the chemical therapy of a patient. |
