PURPOSE:To produce a novel alkali-resistant cellulase capable of maintaining a high activity in an alkaline region and even in comparison with the highest activity in the optimal pH, and useful for detergents for clothing. CONSTITUTION:An alkali-resistant cellulase K-344 is obtained by culturing Bacillus sp. KSM-344 (FERM P-9008) and has following properties: (1) has activities against CMC, cellulose powder, avicel, filter paper and p-nitrophenyl cellobioside, (2) has the optimal pH of 6 and a relative activity in a pH range of 4.5-11 in a quantity of 50% of the activity at the optimal pH, (3) is not deactivated at a pH range of 5-10 and has the activity of >=50% also at a pH range or 4.5-12, when held at 30 deg.C for one hour, (4) has the optimal action temperature at 70 deg.C, (5) has an activity-halving temperature of approximately 55 deg.C, when treated at the optimal pH for 30min, (6) has a mol.wt. of approximately 16,000 by a gel filtration, and (7) is substantially not affected with surfactants, proteases, chelating agents, etc. |
