PURPOSE: To obtain a proliferation-responsive element, having a specific base sequence, present on a promoter of a human cyclin A gene and applicable to the expression of proliferation responsiveness of a specific gene, regulation, etc., of the proliferation rate of a cultured cell for producing a substance. CONSTITUTION: This new proliferation-responsive element has a base sequence represented by the formula and is present on a promoter of a human cyclin A gene and applicable to basic experiments including the expression of a specific gene into the proliferation responsiveness or regulation, etc., of the proliferation rate of a cultured cell used for producing a substance. The element is obtained by screening a genomic DNA library derived from a human placenta according to a hybridization method using a cDNA of the human cyclin A as a probe, obtaining a promoter region of the human cyclin A gene, preparing DNA fragments cleaved from the 5'-terminal thereof in small portions with a nuclease, etc., joining each of the fragments to the upstream side of a reporter gene, expressing each thereof and analyzing the expressed genes. |
