PURPOSE: To easily determine the species of blue mussels useful as an important food resource and necessary from the viewpoint of resource management, breeding, etc., by using the difference of base sequence in a variable region of an adhesive protein gene of blue mussels. CONSTITUTION: Three species of blue mussels (e.g. mussel) i.e., Mytilus-edulis, Mytilus-galloprovincialis and Mytilus-trossulus are discriminated by collecting the gill of the mussels, decomposing the tissue with an enzyme, etc., extracting the DNA by conventional method, amplifying the extract by PCR method using a primer composed of a sense chain at the upstream side of the variable region of the adhesive protein gene of blue mussel and an antisence chain at the downstream side of the variable region, subjecting the amplified product to agarose gel electrophoresis and easily differentiating the three species by the length of the amplified fragment. |
