PURPOSE: To provide the subject new manifestation vector having a DNA to code a human somatostatin receptor protein sub-type 2 and capable of producing a protein useful for the selection of agent for the prevention and treatment of acromegaly, gigantism, gastric ulcer, dwarfism, diabetes, etc. CONSTITUTION: This new manifestation vector pAC01 holding a DNA to code a human somatostatin receptor protein sub-type 2 is produced by synthesizing a DNA oligomer pair consisting of a part of a cDNA base sequence based on the cDNA base sequence of a human somatostatin receptor protein sub-type 2 (SSTR2), carrying out a polymerase chain reaction using the synthesized oligomer pair as a primer and a human pituitary gland cDNA as a template, cloning the cDNA of the human somatostatin receptor protein sub-type 2 and integrating the product in a manifestation vector. A recombinant human somatostatin receptor protein is produced by inserting the obtained manifestation vector pAC01 into a CHO cell deficient in a dihydrofolic acid reductase gene and manifesting the gene. |
