PURPOSE: To obtain a separating agent, containing a polysaccharide derivative, excellent in stability of a base line in operating a column without eluting the polysaccharide derivative of a low molecular weight, having a wide range of solvents usable for an eluent and useful for optical resolution of a racemic modification. CONSTITUTION: This separating agent contains a polysaccharide derivative having >=5 average polymerization degree of the polysaccharide (the average number of pyranose or furanose rings contained in one molecule), 1000-500000 weight-average molecular weight of the polysaccharide derivative (expressed in terms of polystyrene) and 1-3 value of Mw/Mn (Mw is the weight-average molecular weight; Mn is the number-average molecular weight, expressed in terms of the polystyrene) indicating the extent of the molecular weight distribution. Furthermore, the polysaccharide derivative is preferably supported on a carrier (e.g. silica gel) having 1μm to 10mm grain diameter and 10Åto 100μm pore diameter. The polysaccharide derivative is obtained by binding hydroxyl groups in the polysaccharide to a compound having a functional group reactive with the hydroxyl groups through well-known ester bonds, urethane bonds, etc., and converting the polysaccharide into the derivative, and an ester or a carbamate derivative is cited.
