VERFAHREN ZUM KLASSIFIZIEREN VON LEUKOZYTEN UND REAGENZIEN

A reagent for the classification and counting of leucocytes, contains a surfactant of formula (I) and/or a surfactant of formula (II): (where R1 is 10-22C alkyl, alkenyl or alkynyl; R2 is O, -COO- or a group of formula (III); n is 8-30; X is -SO3Na, -COONa, -OSO2ONa or -ONa) and pref. also contains an osmotic pressure raising agent and a solubilising agent. By use of different solubilising agents the leucocytes can be classified into five groups of lymphocytes, monocytes and granulocytes. The solubilising agents for the five groups are: (1) urea, thiourea, N,N-dimethylurea, N,N'-dimethylurea, ethyleneurea, methylurethane, or urethane; (2) octyl beta-D-glucoside, CHAPS, CHAPSO, MEGA-8, MEGA-9, MEGA-10, sucrose monocaproate, or N-formylmethyl leucylalanine; (3) guanidine thiocyanate, hydrochloride, nitrate, rhodanide, carbonate, sulphate or phosphate or guanidylguanidine or 1,1,3,3-tetraguainidie; (4) sodium deoxycholate, cholic acid or taurocholic acid; (5) sodium mono-, di- or trichloroacetate or mono-, di- or tribromoacetate.
