Liquids are treated without contamination in a vessel with a liquid inlet opening (A10) and a liquid outlet opening (A11). The outlet is closed by a component (A12) which presses elastically against the opening and can be reversibly opened by reduced or elevated pressure. The component pref. exerts a permanent force in the direction of the outlet opening. The vessel is pref. hollow cylindrical and may have a slightly tapering outside shape with a precisely cylindrical interior. Also claimed is a method for isolating a substance from a sample which is put in the vessel by adding a matrix which can immobilise the substance and incubating for a period sufficient for immobilisation before removal of liquid through the outlet opening.
